Studies on the Lectin Binding Patterns of the Oviduct Luminal Epithelium and Oocytes in Mouse




Abstract
In order to study the gametes interactions which mediated by cell surface recognition molecules bearing complementary carbohydrate residues, and to examine interaction between oocytes and oviduct, sugar residues on extracellular matrix and
membrane
surface of mouse oocytes and oviduct tissue were examined using two kinds of lectins conjugated with TRITC in toto. Cumulus cell surface was strongly labeled with LCA but sparsely with UEA-I. LCA bound to zona pellucida and perivitellin space of
oocyte
after ovulation and onward. UEA-I bound to same sites after fertilization. Once fertilization had occured, oolemma was bound by both LCA and UEA-I. Lectin binding to oolemma were more prominant when exmined in whole mounted but not detected in
paraffin
embedded specimens. Luminal epithelium of oviduct was strongly labeled with LCA but not with UEA-I. This result suggested that mannose residues were present in zona pellucida and perivitelline space of oocyte, and luminal epithelium of oviduct.
Mannose
residues were rich in ZP and appear in oolemma after fertilization. Both fucose and mannose residues were rich in cumulus cells surface and cumulus matrix. There was no difference in the distribution of mannose residues on the ZP and PVS between
fertilized oocytes and aged unfertilized oocytes. These results suggest that perivitelline space of the oocyte is not an empty space and substances having mannose residues originated from oviduct might interact with oocyte after ovulation.
